THE CHROMOSOME END REPLICATION: LESSONS FROM MITOCHONDRIAL GENETICS Jozef Nosek1, ubomr Tomska2 and Blanka Kucejov1 1 Department of Biochemistry, Faculty of Natural Sciences, Comenius University, Bratislava, Slovak republic 2 Department of Genetics, Faculty of Natural Sciences, Comenius University, Bratislava, Slovak republic The widespread occurrence of linear mitochondrial genomes evokes intriguing questions concerning the evolutionary origin and mechanisms leading to the emergence and stabilization of linear DNA genophores. The study of their replication strategies opens a unique possibility to discover alternative solutions to the endreplication problem and to elucidate how these mechanisms have appeared in evolution. The analysis of linear mitochondrial genomes in organisms belonging to different phylogenetic lines indicates that their evolutionary emergence was accompanied by the generation of various types of terminal structures, the adaptation of existing replication machinery and by the application of different strategies of the telomere replication. This scenario is illustrated by the molecular anatomy and replication of the linear mitochondrial genome in the opportunist yeast pathogen Candida parapsilosis. Recent studies have revealed the existence of extragenomic minicircular molecules derived from the telomere repeats that seem to participate in the novel pathway of telomere maintenance. Importantly, several lines of evidence indicate that a similar mechanism may also be involved in the alternative, telomerase-independent, maintenance of nuclear telomeres in higher eukaryotes, including human telomerase-negative tumor cells.
Calls to extend a competition inquiry beyond the market for groceries to include, for example, pharmaceuticals, have been rejected by the Competition Commission. A statement of issues released by the commission last week says that the commission will not extend its investigation beyond the defined remit of groceries, which means food, pet foods, drinks, cleaning products, toiletries and household goods. However, the statement adds that the Enterprise Act 2002 gives the commission power to identify features that affect competition in any market in which grocery suppliers operate. But it can only take account of matters such as the mix of shops on high streets and its impact on local communities if they adversely affect competition by leading to higher prices, less choice, lower quality of available products or less innovation. In this context, the issues statement says: "We will consider . whether the activities of grocery retailers affect competition in markets for non-grocery items, such as [pharmaceuticals]." Inquiry chairman Peter Freeman said: "We know there are many issues of more general public concern surrounding the grocery market.We will listen to evidence on these, but our concern must be with their impact on competition; we hope those who provide evidence to us will focus on that." Thus, Hin recombines pREC and knots pKNOT. Although we initially anticipated that pREC would serve to differentiate between effects caused by recombination and those caused by DNA knotting, we data not shown ; and others observed that, in vivo, Hin will occasionally mediate processive recombination events to knot plasmids containing wild-type recombination sites at a steady-state level of 2 3% Table 1 ; [25]. pKNOT is extensively knotted by Hin because the mismatch between the sites requires processive recombination [21, 25, 26] for religation. Hin expression increases the steady-state knotting of pKNOT approximately 5- to 10-fold over endogenous levels in the presence of topoisomerase IV function and 35to 45-fold when topoisomerase IV function is blocked [21, 25]. Twist knots with three 31 ; , five 52 ; and seven 72 ; negative nodes as well as composite knots with six 31#31 ; , eight 31#52 ; and nine 31#31#31 ; nodes are readily generated when Hin knots pKNOT [25]. Higher noded knots are seen in decreasing abundance. Only twist knots with three 31 ; and four 41 ; nodes are seen when Hin knots a plasmid with two wild-type recognition sites [25]. Previous studies examined the effect of Hin and other site-specific recombinases on gene expression [32, 33]. However, a key distinction between those studies and the experiments performed here is that in those experiments the recombinase binding sites were placed between the promoter and the gene whereas here the reporter gene is distant to the site of recombination. Thus, here we examine the global effects on the DNA molecule rather than the local effects on promoter function and imdur.

FINDING OF NO SIGNIFICANT IMPACT SECTION 26A AND LAND USE APPROVAL FOR NUCOR STEEL DECATUR, LLC PORT FACILITY EXPANSION AT TENNESSEE RIVER MILE 297.5, LEFT BANK, WHEELER RESERVOIR, MORGAN COUNTY, ALABAMA Nucor Steel Decatur, LLC proposes to expand its existing port facility along the Finley Island back channel adjacent to the Mallard-Fox Creek Industrial Park. Nucor has requested TVA approval of the facility expansion under Section 26a of the TVA Act and that TVA grant it the rights to use 12.4 acres of submerged TVA-owned land for the facility expansion. The port expansion would allow Nucor to expand its business by delivering an increased volume of raw materials by barge. The proposed port facility expansion also requires approval by the U.S. Army Corps of Engineers Corps ; under Section 10 of the Rivers and Harbor Act and under Section 404 of the Clean Water Act. TVA has cooperated with the Corps in the preparation of an environmental assessment EA ; of the proposed facility expansion. TVA is adopting this EA and it is incorporated by reference. The EA evaluates three alternatives: 1 ; No Action, under which the permit requests and approvals would be denied; 2 ; Applicant's Proposed Action; and 3 ; Applicant's Proposed Action with Special Conditions. The applicant's proposed action consists of dredging and disposing of about 121, 000 cubic yards of fill, installing a total of six mooring cells in two areas to allow the fleeting of 71 additional barges, extending the length of the existing dock from 350 to 890 feet, and constructing a new scrap barge unloading dock. The Corps proposes to approve Alternative 3. The Nucor plant is in an existing industrial park which was the subject of an Environmental Impact Statement prepared by TVA in 1979. The existing Nucor plant and port facility were originally built by Teicor Steel Company, LLC, and were the subject of an EA prepared by TVA in 1995. The subject EA concludes that impacts to air quality, water quality, aquatic life, wildlife, vegetation, recreation, aesthetics, land-based transportation, and navigation would be minor and insignificant.
